    目的 探讨联合应用PI3K 和mTOR 双重靶向抑制剂BEZ235 及上皮间质转换(EMT)抑制 剂AZD6244 对人宫颈癌细胞增殖、迁移能力的影响。方法 采用MTT 法分析BEZ235、AZD6244 对Hela 和SiHa 细胞增殖能力的影响,损伤愈合实验检测BEZ235、AZD6244 对Hela和SiHa 细胞迁移能力的影响, Western blot 检测BEZ235、AZD6244 对EMT 相关标志物蛋白的表达变化。结果 BEZ235、AZD6244 对宫 颈癌细胞Hela 和SiHa 的增殖有协同抑制作用;联合应用BEZ235、AZD6244 可以通过EMT 途径抑制Hela 和SiHa 细胞的侵袭、迁移。结论 BEZ235、AZD6244 联合应用可体外抑制Hela 和SiHa 细胞的增殖、迁移, 其部分机制是通过抑制EMT 途径实现的。

    Abstract:

    Objective To detect the effect of co-treatment with BEZ235 (inhibitor of PI3K and mTOR) and AZD6244 (inhibitor of EMT) on the proliferation and migration of human cervical carcinoma cell lines Hela and Siha. Methods Cell proliferation ability was detected by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-Htetrazolium bromide (MTT) assay, and the capacity of cell migration was measured by wound healing assay. Western blot was used to detect the changes of the expressions of EMT-related proteins. Results The proliferation of Hela and SiHa cells was effectively inhibited by co-treatment with BEZ235 and AZD6244. In addition, the combined treatment also synergically suppressed the invasion and migration of Hela and SiHa cells through EMT pathway. Conclusions Combined treatment with BEZ235 and AZD6244 may synergically inhibit the proloferation and migration of cervical cancer cells in vitro , and part of the mechanism may be completed by weakening the EMT pathway.
