Abstract:Objective To investigate the effect of Schisanhenol (Sal) on proliferation of cardiac fibroblasts (CFb) and potential mechanisms. Methods Purified CFb were obtained by standard methods (digestive enzyme and differential adhesion). CFb were induced by angiotensin II (Ang II) and identified by immunofluorescence. Cellular proliferation of CFb was measured by methyl thiazolyl tetrazolium (MTT) assay. Levels of TGF-β1, collagen I and collagen III proteins were examined by Western blotting. Results Sal inhibited proliferation of CFb in a dose-dependent manner (P < 0.05). Expression of collage I, collagen III, and TGF-β1 induced by Ang II was downregulated by treatment of Sal in a dose-dependent manner (P < 0.05). Conclusion Sal inhibits cellular proliferation and expression of collagen through inhibition of the TGF-β1.
