Abstract:Objective To study the expression of interleukin-22 (IL-22) in mice with systemic lupus erythematosus(SLE), and explore the significance of IL-22 in the pathogenesis of SLE. Methods Eighty mice were randomly divided into SLE group (40 cases) and control group (40 cases). The mice of the SLE group were injected with activated splenocytes through caudal vein to establish SLE mouse model. Those of the control group were injected with the same amount of normal saline through caudal vein. In the 4th and 8th weeks after model establishment, the kidney morphology on HE sections, the titer of serum anti-double stranded DNA antibody, serum IL-22 level, the expressions of IL-22 mRNA and protein in the kidney and lymphoid tissues were compared between the two groups.Results Histological study revealed degeneration of renal tubular epithelial cells, a large number of inflammatory cell infiltration in the renal interstitium, and proliferation of mesangial cells in the SLE group; but normal renal morphology in the control group. In the 4th and 8th weeks after modeling, the serum titer of anti-double stranded DNA antibody in the SLE group was higher than that in the control group (P < 0.05); the serum level of IL-22 in the SLE group was not statistically different from that in the control group (P > 0.05); the expression of IL-22 mRNA in the kidney tissues of the SLE group was higher than that of the control group (P < 0.05); but the expression of IL-22 mRNA in the lymphoid tissues of the SLE group was not statistically different from that of the control group (P >0.05). The expression of IL-22 protein in the kidneys of the SLE group was strongly positive and concentrated in the glomeruli, while IL-22 protein was not obviously expressed in the kidneys of the control group. IL-22 protein was not expressed in the lymphoid tissues of either group. Conclusions The systemic lupus erythematosus mice have the pathological changes of systemic lupus erythematosus in the kidneys. The expressions of IL-22 mRNA and protein are increased in the renal tissues.
