Abstract:Objective To investigate the expression of T-cell immunoglobulin domain and mucin domain-1 (TIM- 1) in rats with IgA nephropathy (IgAN), and to explore the role of TIM-1 in the pathogenesis of IgAN. Methods Twenty male SD rats were randomized into two groups, i.e. IgAN model group (10 rats) and control group (10 rats).The rats were sacrificed after the completion of building the model and the kidneys were obtained. The pathological changes of the rat kidneys were observed after PAS and immunofluorescence staining. Immunohistochemical staining was used to detect the expression of TIM-1 protein and RT-PCR was used to measure the expression of TIM-1 mRNA in the renal tissue. Results TIM-1 expression in the IgAN group mainly distributed in glomeruli and renal tubules, the expression levels of TIM-1 mRNA and protein significantly increased in the IgAN group compared with those in the control group (p < 0.05 ). A positive correlation was found between the expression quantity of TIM-1 mRNA and Katafuchi score (r = 0.846,p < 0.05 ). Conclusions TIM-1 may be involved in the occurrence and development of IgAN.
