灯盏花素对肝癌细胞 HepG2 和结肠癌细胞 Caco2 的体外抗肿瘤活性筛选

摘要:

观察灯盏花素对人肝癌细胞（HepG2）和人结肠癌细胞（Caco2）的体外增殖抑制影响。方法以不同浓度的灯盏花素分别处理 HepG2 和 Caco2 细胞 24、48和 72 h 后采用 MTT 法检测灯盏花素对 2 种细胞活力的影响，以半数抑制浓度（IC50）及治疗指数（TI）作为评价其抗肿瘤活性及安全性指标。结果 灯盏花素对 HepG2 表现高浓度抑制作用，浓度为 20 000μmol/L 时，对 HepG2 细胞 24、48和 72 h 的抑制率分别为37%、63%和 69%；对 Caco2 细胞呈浓度依赖性，浓度为 5 000μmol/L时，对 Caco2 细胞 24、48和 72 h 的抑制率分别为72%、95%和 92%。灯盏花素对 HepG2 24h的 IC50 值为 24320.5μmol/L；48h的 IC50 值为 30 667.7μmol/L；72 h 的 IC50 值为 15 586.1μmol/L。对 Caco2 细胞 24 h 的 IC50 值为 11 417.0μmol/L；48 h 的 IC50 值为832.2μmol/L；72 h 的 IC50 值为 1 369.2μmol/L。结论 灯盏花素对 HepG2 和 Caco2 细胞均有不同程度的抑制作用，对 Caco2 的抑制效果更明显。

Abstract:

To investigate the inhibitory effects ofbreviscapine on proliferation of human hepatocellularcarcinoma cellline HepG2 and colon cancercellline Caco2.M ethodsBoth HepG2 and Caco2 cell lines were treated with breviscapine atdifferentconcentrations for24,48 and 72 h,and the inhibitory rate was analyzed by MTT assay.The 50% inhibitory concentration (IC50) and the therapeutic index (TI) were used as indicators to assess their anti-tumor activity and safety. Results Breviscapine at high concentration displayed inhibitoryeffecton HepG2;when theconcentration was20,000 μmol,itsinhibitoryrateto HepG2 cellsafter24,48 and 72 h was37% ,63% and 69% respectively.Asto Caco2 cellsbreviscapine showed concentration dependency; when the concentration was 5,000 μmol,its inhibitory rate after 24,48 and 72 h was 72% , 95% and 92% respectively.The IC50 ofbreviscapine on HepG2 cellsafter24,48 and 72 h was 24,320.5 μmol,30,667.7 μmol and 15,586.1 μmolrespectively；while thaton Caco2 cellsafter24,48 and 72 h was11,417.0 μmol,832.2 μmol and 1,369.2 μmolrespectively.Conclusions Breviscapine exhibits differentdegree ofinhibitory effecton HepG2 and Caco2 cells,theeffectismoresignificanton Caco2 cells.
