    摘要:

    探讨美洲大蠊提取物体外对肝癌(HCC)HepG2细胞增殖、迁移能力,以及细胞中血管内皮生长因子(VEGF)蛋白表达的影响。方法噻唑蓝(MTT)法检测细胞增殖;划痕实验检测美洲大蠊提取物对HCC细胞迁移能力的影响;免疫细胞染色法、酶联免疫吸附试验(ELISA)检测美洲大蠊提取物对细胞中VEGF蛋白表达的影响。结果MTT和划痕实验结果表明,美洲大蠊多肽、CⅡ-3和脱脂膏能抑制人HCC HepG2细胞的增殖和迁移,且呈时间、剂量依赖关系;免疫细胞化学染色法和ELISA 法结果显示,美洲大蠊多肽、CⅡ-3及脱脂膏能下调人HCC HepG2细胞中VEGF 蛋白的表达。结论美洲大蠊多肽、CⅡ-3 及脱脂膏能抑制人HCC HepG2细胞增殖和迁移,同时能降低HepG2细胞中VEGF蛋白的表达,且美洲大蠊多肽的作用优于CⅡ-3 和脱脂膏。

    Abstract:

    To investigate the effect of extracts from on proliferation,migration and VEGF protein expression of human hepatocellular carcinoma HepG2 cells. Methods Scratch assay was applied to measure the effect of extracts from on migration of HepG2 cells. Immunohistochemical staining and enzyme-linked immunosorbent assay (ELISA) were used to detect the effect of extracts from on the expression of VEGF protein in the cells. Results MTT assay and Scratch assay showed that polypeptides, CⅡ-3 and skimmed cream could inhibit the proliferation and migration of HepG2 cells in a time- and dose-dependent manner. Immunohistochemical staining and ELISA indicated that polypeptides, CⅡ-3 and skimmed cream could down regulate the expression of VEGF protein in the cells. Conclusions polypeptides, CⅡ-3and skimmed cream can inhibit the proliferation and migration of HepG2 cells in a timeand dose-dependent way, and reduce the expression of VEGF protein in the cells; and the effect of extracts is better than that of CⅡ-3 and skimmed cream.
