    摘要:

    目的  探讨中国汉族人群肿瘤坏死因子-α(TNF-α)基因单核苷酸多态性(rs1800629和rs361525)与乙型肝炎病毒(HBV)感染的相关性。方法  用TaqMan探针对纳入的1 097例样本进行TNF-α、rs1800629和rs361525位点多态性检测,血样本包括429例慢性HBV感染者(CH组)、429例乙肝感染后自发清除者(SR组)及239例健康对照者(HC组)。结果  ①CH组与SR组比较,rs1800629 GA基因型与G等位基因与感染后病毒的自发清除有关[基因型:■=0.369,P =0.000;等位基因:■=2.210,P =0.000;显性模型(GG vs GA+AA):■=0.536,P =0.000]。②CH组与SR组比较,SNP rs361525 GA基因型或A等位基因与感染后病毒的自发清除密切相关[基因型:■=0.288,P =0.000;等位基因:■=0.549,P =0.000;显性模型(GG vs GA+AA):■=0.634,P =0.000]。结论  TNF-α基因多态性与中国汉族人群HBV病毒感染后的自发清除密切相关。

    Abstract:

    Objective To assess the relation between two single nucleotide polymorphisms (rs1800629 and rs361525) of TNF-α gene and the outcomes of HBV infection in Chinese Han population. Methods TNF-α (rs1800629 and rs361525) was genotyped by TaqMan probe method in 1,097 subjects, including 429 patients with chronic HBV infection (CH), 429 HBV spontaneous clearance subjects (SR) and 239 healthy control individuals (HC). Results rs1800629 GA genotype and G allele showed significant protective effect on HBV spontaneous clearance [CH vs SR: genotype: ■ = 0.369, P = 0.000; allele: ■ = 2.210, P = 0.000; dominant model (GG vs GA+AA): ■ = 0.536, P = 0.000]. SNP rs361525 showed a significant association with HBV spontaneous clearance. The rs361525 GA genotype and A allele played a protective role [CH vs SR: genotype: ■ = 0.288, P = 0.000; allele: ■ = 0.549, P = 0.000; dominant model (GG vs GA+AA): ■ = 0.634, P = 0.000]. Conclusions TNF-α gene polymorphisms are associated with HBV spontaneous clearance.
