白细胞介素-37在结肠癌组织中的表达及其对结肠癌细胞增殖、侵袭的作用

Expression of IL-37 in colon cancer and its inhibition on proliferation and invasion of colon cancer cells in vitro

摘要:

目的 探讨白细胞介素-37（IL-37）在结肠癌组织中表达的变化，并通过体外实验探讨其对结肠癌细胞增殖、侵袭的作用。方法 收集2017年4月—2019年4月在西安市第四医院手术切除的结肠癌组织及对应的癌旁组织，检测其IL-37的表达量；培养结肠癌HT29细胞，随机分为对照组、转染空白pcDNA3.1质粒的空白质粒组、转染表达IL-37的pcDNA3.1重组质粒的IL-37质粒组，MTS检测3组结肠癌HT29细胞的增殖能力，Transwell实验检测细胞的侵袭能力，qRT-PCR检测细胞周期蛋白D1（CyclinD1）、B淋巴细胞瘤-2（Bcl-2）、基质金属蛋白酶2（MMP-2）、基质金属蛋白酶9（MMP-9）mRNA相对表达量，Western blotting法检测IL-37、p-STAT3及p-Akt蛋白相对表达量。结果 结肠癌组织中IL-37的蛋白相对表达量低于癌旁组织（P <0.05）；3组细胞增殖及侵袭能力，CyclinD1、Bcl-2、MMP-2、MMP-9 mRNA相对表达量，p-STAT3、p-Akt的蛋白相对表达量比较，差异有统计学意义（P <0.05），IL-37质粒组的细胞增殖及侵袭能力，CyclinD1、Bcl-2、MMP-2、MMP-9 mRNA相对表达量，p-STAT3、p-AKT的蛋白相对表达量均低于对照组、空白质粒组。结论 IL-37在结肠癌组织中表达减少，上调IL-37的表达能够抑制结肠癌细胞的增殖、侵袭，该抑制作用可能与抑制STAT3/Akt通路有关。

Abstract:

Objective To investigate the expression of interleukin-37 (IL-37) in colon cancer and its inhibition on the proliferation and invasion of colon cancer cells.Methods The colon cancer tissues and the corresponding adjacent tissues resected in our hospital during April 2017 to April 2019 were collected, then the expression of IL-37 was detected. The colon cancer HT29 cells were cultured and randomly divided into control group, blank plasmid group transfected with blank pcDNA3.1 plasmid, and IL-37 plasmid group transfected with pcDNA3.1 recombinant plasmid expressing IL-37. The proliferation and invasion activity, the mRNA expression of proliferation and invasion genes (CyclinD1, Bcl-2, MMP-2, MMP-9), the protein expression of p-STAT3 and p-Akt were determined.Results The protein expression of IL-37 in colon cancer was lower than that in adjacent tissues (P < 0.05); the difference of the cell proliferation and invasion activity, the mRNA expression of CyclinD1, Bcl-2, MMP-2, and MMP-9, and the protein expression of p-STAT3 and p-Akt among 3 groups had statistical significance (P < 0.05); the cell proliferation and invasion activity, the mRNA expression of CyclinD1, Bcl-2, MMP-2, and MMP-9, and the protein expression of p-STAT3 and p-Akt in IL-37 plasmid group were significantly lower than those in control group and blank granulation group (P < 0.05).Conclusion The expression of IL-37 is decreased in colon cancer, and the increase of IL-37 expression can inhibit the proliferation and invasion of colon cancer cells, which is related to the inhibition of STAT3/Akt pathway.
