结直肠癌患者血清microRNA-335、microRNA-497表达与临床病理特征及预后的关系

Relationship of serum miR-335 and miR-497 expression with clinicopathological features and prognosis in patients with colorectal cancer

目的 探究结直肠癌患者血清microRNA-335（miR-335）、microRNA-497（miR-497）表达与临床病理特征及预后的关系。方法 选取2013年2月—2014年9月于邯郸市中心医院诊治的99例结直肠癌患者作为结直肠癌组，选择同期在该院体检的99例健康者作为健康组。采用实时荧光定量PCR（qRT-PCR）检测两组的血清miR-335、miR-497表达水平，比较不同临床病理特征患者血清miR-335、miR-497的表达水平，采用Kaplan-Meier法估计不同血清miR-335、miR-497表达水平患者的生存率。结果 结直肠癌组患者血清miR-335、miR-497表达水平低于健康组（P <0.05）；浸润深度T₁、T₂，TNM分期Ⅰ、Ⅱ，及无淋巴结转移患者血清miR-497表达水平高于浸润深度T₃、T₄，TNM分期Ⅲ，及有淋巴结转移患者（P <0.05）；TNM分期Ⅰ、Ⅱ和中高分化患者血清miR-355表达水平高于TNM分期Ⅲ和低分化患者（P <0.05）；血清miR-335和miR-497高表达的结直肠癌患者5年生存率分别高于血清miR-497和miR-335低表达患者（P <0.05）。结论 结直肠癌患者血清miR-335、miR-497表达水平下降与结直肠癌的发生、发展，病情恶化及预后不良有关。

Abstract:

Objective To investigate the relationship of serum Microrna-335 (miR-335) and miR-497 expression in colorectal cancer patients with their clinicopathological characteristics and prognosis.Methods From February 2013 to September 2014, 99 colorectal cancer patients who were diagnosed and treated in our hospital were selected as the colorectal cancer group, and 99 healthy patients who underwent physical examination in our hospital during the same period were selected as the healthy group. Fluorescence quantitative PCR was used to detect the expression levels of serum miR-335 and miR-497 in the two groups, and the clinicopathological characteristics of patients with different serum miR-335 and miR-497 expression levels were compared. Kaplan-Meier method was used to analyze the prognosis of patients with different serum miR-335 and miR-497 expression levels.Results The expression levels of serum miR-335 and miR-497 in the colorectal cancer group were lower than those in the healthy group (P < 0.05). The expression level of serum miR-497 in patients with infiltrative depth T₁-T₂, TNM stage I/II, and without lymph node metastasis were higher than those in patients with infiltrative depth T₃-T₄, TNM stage III, and with lymph node metastasis (P < 0.05). The expression level of serum miR-335 in patients with TNM stage I/II, Medium and high differentiation were higher than than those in patients with TNM stage III and low differentiation (P < 0.05). 5-year survival rates of colorectal cancer patients with high serum miR-497 expression and high miR-335 expression were higher than those with low serum miR-497 expression and low miR-335 expression respectively (P < 0.05).Conclusion The decreased expression levels of serum miR-335 and miR-497 in colorectal cancer patients were correlated with the occurrence, development, deterioration, and poor prognosis of colorectal cancer.
