    摘要:

    目的 探讨薏苡仁提取物(CSE)对糖尿病肾病大鼠肾脏功能的保护作用及相关机制。方法 对大鼠腹腔注射小剂量链脲佐菌(STZ)复制糖尿病肾病模型。采取不同的干预措施,将50只大鼠随机分为5组:对照组(ND组)、链脲佐菌模型组(STZ组)、格列齐特治疗组(GLI组)、CSE低剂量组(CSE-L组),CSE高剂量组(CSE-H组)。检测各组大鼠空腹血糖水平、24 h尿蛋白含量、初始体重、最终体重、肾脏重量及肾组织损伤指标。Western blotting法检测肾组织中转化生长因子-β1(TGF-β1)和髓样分化因子88(MyD88)蛋白的相对表达量。结果 各组大鼠不同时间的空腹血糖水平差异有统计学意义(P <0.05),6周时血糖低于1周和3周时血糖;各组大鼠的空腹血糖水平差异有统计学意义(P <0.05),STZ组高于ND组(P <0.05),CSE-L组、CSE-H组低于STZ组(P <0.05)。各组大鼠空腹血糖水平的变化趋势差异有统计学意义(P <0.05)。各组大鼠24 h尿蛋白含量、初始体重、最终体重比较,差异有统计学意义(P <0.05),STZ组24 h尿蛋白水平高于ND组(P <0.05),初始体重、最终体重低于ND组(P <0.05),CSE-L组、CSE-H组24 h尿蛋白水平低于STZ组(P <0.05),初始体重、最终体重高于STZ组(P <0.05)。各组大鼠肾功能参数比较,差异有统计学意义(P <0.05),STZ组Scr、BUN、TC、HDL-C及IL-6水平高于ND组(P <0.05);CSE-H组Scr、BUN、TG、TC、HDL-C、GSH、IL-6水平低于STZ组(P <0.05)。各组大鼠肾组织中TGF-β1和MyD88蛋白相对表达量比较,差异有统计学意义(P <0.05),STZ组肾组织中TGF-β1蛋白相对表达量高于ND组(P <0.05),MyD88蛋白相对表达量低于ND组(P <0.05),CSE-H组肾组织中TGF-β1蛋白相对表达量低于STZ组(P <0.05),MyD88蛋白相对表达量高于STZ组(P <0.05)。结论 CSE可通过调节TGF-β1、MyD88抑制糖尿病肾病大鼠肾脏纤维化进程和炎症反应,具有良好的肾脏保护作用。

    Abstract:

    Objective To investigate the protective effect of CSE on renal function in DN rats and its mechanism.Methods Fifty rats were randomly divided into ND group, STZ group, Gli group, CSE-L group, and CSE-H group. Fasting blood glucose level, 24-hour urinary protein content, initial body weight, final body weight, renal mass and renal tissue injury indexes were measured. The expression of MyD88 protein and TGF-β in renal tissue were detected.Results The fasting blood glucose levels of rats in each group were compared at different time points, 6 w was lower than 1 w and 3 w, and the difference was statistically significant (P < 0.05); that in STZ group was higher than in ND group (P < 0.05); that in CSE-L group and CSE-H group were higher than in STZ group. The 24 h urine protein content, initial weight, and final weight of the rats in each group were compared with statistical significance (P < 0.05); the urine protein level of the STZ group was higher than that of the ND group, and the initial and final weight were lower than the ND group (P < 0.05). CSE-L group and CSE-H group had lower urine protein levels than STZ group, and initial body weight and final body in CSE-L group and CSE-H group weight were higher than in STZ group (P < 0.05). The comparison of the renal function parameters of rats in each group showed statistically significant differences (P < 0.05); the levels of Scr, BUN, TC, HDL-C, and IL-6 in the STZ group were higher than those in the ND group (P < 0.05); the levels of Scr, BUN, TG, TC, HDL-C, GSH and IL-6 in CSE-H group were lower than those in STZ group (P < 0.05). The expression of TGF-β1 and MyD88 in the kidney tissue of rats in each group was compared, and the difference was statistically significant (P < 0.05). The expression level of TGF-β1 protein in the kidney tissue of the STZ group was higher than that of the ND group (P < 0.05); the protein expression level of MyD88 was lower than that of the ND group (P < 0.05); the TGF-β1 protein expression level in the kidney tissue of the CSE-H group was lower than that of the STZ group (P < 0.05), and the protein expression level of MyD88 was higher than that of the STZ group (P < 0.05).Conclusion CSE can inhibit the process of renal fibrosis and inflammatory reaction in diabetic nephropathy rats with a good renal protective effect, which has potential application value in the clinical treatment of diabetic nephropathy.
