Abstract:Objective To investigate the expression of talin-1 (Talin1) in the placentas of patients with preeclampsia and its mechanism of influence on the biological behavior of trophoblast cells. Methods Placental tissues were collected from 22 healthy pregnant women and 20 patients with preeclampsia. The expression of Talin1 in the two groups was analyzed by Western blotting. Human trophoblast cells (HTR8/Svneo cells) were divided into six groups: pcDNA3.1 NC group, pcDNA3.1 Talin1 group, siRNA NC group, siRNA Talin1 group, agonist group, and siRNA Talin1 + inhibitor group. The proliferation of HTR8/Svneo cells was detected by MTT assay; the invasion and migration of HTR8/Svneo cells were detected by Transwell assay; the apoptosis of HTR8/Svneo cells was detected by flow cytometry; and the expression of Talin1 and Wnt/β-catenin pathway-related proteins in HTR8/Svneo cells was analyzed by Western blotting. Results The expression of Talin1 in the placentas of patients with preeclampsia was increased (P < 0.05). Compared with the pcDNA3.1 NC group, the expression of Talin1 in the pcDNA3.1 Talin1 group was increased, and the proliferation, invasion, and migration abilities of HTR8/Svneo cells were weakened, the apoptosis rate was increased, and the expression of Wnt 3α and β-catenin proteins was decreased (P < 0.05). Compared with the siRNA NC group, the expression of Talin1 in the siRNA Talin1 group and the siRNA Talin1 + inhibitor group was decreased (P < 0.05). There was no significant difference in the expression of Talin1 between the agonist group and the siRNA NC group (P > 0.05). Compared with the siRNA NC group, the proliferation, invasion, and migration abilities of HTR8/Svneo cells in the siRNA Talin1 and agonist groups were enhanced, the apoptosis rate was decreased, and the expression of wingless-type MMTV integration site family 3α (Wnt 3α) and β-catenin proteins was upregulated (P < 0.05). There was no significant difference in the proliferation, invasion, migration ability, apoptosis rate, and expression of Wnt 3α and β-catenin proteins between the siRNA Talin1 + inhibitor group and the siRNA NC group (P > 0.05).Conclusion The expression of Talin1 is increased in the placentas of preeclampsia patients. Inhibition of Talin1 expression can promote the proliferation, migration and invasion of trophoblast cells and inhibit the apoptosis of trophoblast cells by activating Wnt/β-catenin signaling axis.
